Research Interests

Our group is interested in understanding gene control by Ets proteins. We have focused our studies on the Erg protein functions during skeletal, mainly cartilage, formation. Recent works have shown that the Erg transcription factors are involved in diverse diseases including leukemia and prostate cancer. Since the prostate cancer metastases clearly have a propensity for bone, we are currently studying the potential link between ERG expression, skeletal development and bone metastases.

Thème de recherche

Basé à l'Institut de Biologie de Lille, notre groupe de recherche s'intéresse à la régulation transcriptionnelle et aux protéines Ets. Nous étudions particulièrement le rôle des protéines Erg dans la mise en place du cartilage et, plus largement du squelette. L'implication récente du gène Erg dans le cancer de la prostate nous amène à élargir notre étude et à considérer son implication dans les métastases osseuses.

The PEA3 group members in mammary metastasis

Development of epithelial tissues, such as mammary gland, is orchestrated by sequential and reciprocal tissue interactions. An essential event in organogenesis is branching morphogenesis, which is dependent on continuous interactions between an epithelial anlage and the adjacent mesenchyme. A crucial determinant of this phenomenon is the spatially and temporally coordinated expression of tissue-specific transcription factors. The PEA3 group members, which are Ets transcription factors, exert a role in this tissue rearrangement. In cancer tissues, they are over-expressed in many metastatic cancers from epithelial origin such as mammary cancers.

One of the main objectives is now to better define the mechanisms by which these transcription factors regulate mammary cancer metastasis. For this purpose, we have developed “in cellulo” and in vivo models in which these factors are either up- or down-regulated. The transcriptional activity of these factors is regulated by various post-translational modifications in order to regulate transcription of their target genes. The cross-talk between post-translational modifications on Serine/Threonine (Phosphorylation) and on Lysine (Acetylation, Ubiquitination and Sumoylation) within these factors is now precisely studied. This could help us to 1) better understand how these transcription factors regulate their target genes, and 2) more generally to depict the roles of these post-translational modifications in general transcription processes.
